Roche Drug Extends Survival in KRAS Lung Cancer Trial
Roche’s investigational drug divarasib extended median survival and delayed disease progression compared with two approved treatments in a Phase III trial of patients with previously treated advanced lung cancer carrying a KRAS G12C mutation. In the 338-person randomized KRAScendo 1 trial, median overall survival was 17.7 months for patients given divarasib, compared with 12.0 months for those receiving sotorasib or adagrasib. The reported hazard ratio for death was 0.56, corresponding to a 44% lower risk of death during the study period for the divarasib group. Median progression-free survival was 9.8 months with divarasib and 5.7 months with the comparison drugs. The hazard ratio for disease progression or death was 0.58, a 42% reduction. Among 328 participants evaluated for response, confirmed response rates were 58.4% with divarasib and 32.3% with sotorasib or adagrasib. The study results were presented at the 2026 European Society for Medical Oncology Congress and published in The New England Journal of Medicine on October 25. The trial enrolled people with advanced or metastatic non-small cell lung cancer whose disease had progressed after earlier treatment. Participants...
